Separation of anticoagulant and fibrinolytic components from Agkistrodon acutus venom and their effects on the coagulation system

Objective: To study the effect of purified thrombin like enzyme and plasmin from Agkistrodon acutus venom on blood coagulation system

Methods: Thrombin like enzyme and plasmin were isolated and purified from the venom of Agkistrodon acutus by DEAE Sepharose CL-6B and Sephadex G-75 chromatography, and their effects on coagulation system indexes were observed through in vivo experiments. Results: Thrombin like enzyme and plasmin were isolated from the venom of Agkistrodon acutus, and their relative molecular weights were 39300 and 26600, respectively, Both thrombin like enzyme and plasmin from Agkistrodon acutus venom can significantly prolong the whole blood coagulation time, activated partial prothrombin time, thrombin time and prothrombin time, and reduce the content of fibrinogen, but the effect of thrombin like enzyme is stronger, while plasmin shows anticoagulant effect only at a larger dose, and the combination of the two is better than their single use

Conclusion:

Thrombin like enzyme and plasmin from Agkistrodon acutus venom have an effect on the blood coagulation system in animals, and the combination of the two has obvious anticoagulant effect
