Effect of Inhibitory Component I of Agkistrodon halys Venom on Migration Activity of Vascular Endothelial Cells

To observe the effect of the anti-tumor fraction I of Agkistrodon acutus venom (AAVC-1) on the migration activity of human umbilical vein endothelial cells (HUVECs), and to explore the possible mechanism of AAVC – Ⅰ inhibiting angiogenesis. Methods: HUVECs were cultured in vitro with AAVC – Ⅰ (0, 20, 40, 80 μ G/ml) treated cells were incubated for 24 h. Scratch test and chemotactic chamber test were used to observe the effect of AAVC – Ⅰ on endothelial cell migration activity; RT-PCR and Western blot were used to detect the changes of mRNA and protein levels of P-selectin and intercellular adhesion factor (ICAM-1) before and after the drug treatment. Results: Compared with HU VECs in the normal group, the migration ability of cells in AAVC – Ⅰ concentration groups decreased to varying degrees, and the expression of P-selectin and ICAM-1 mRNA decreased significantly. Conclusion: AAVC – Ⅰ may inhibit the migration activity of endothelial cells by down regulating the mRNA and protein levels of P-selectin and ICAM-1
