Separation of anticoagulant and fibrinolytic components from Agkistrodon halys venom and their effects on blood coagulation system

To study the effect of thrombin-like and fibrinolytic enzymes isolated and purified from the same Agkistrodon acutus venom on the blood coagulation system. Methods: The thrombin-like and fibrinolytic enzymes were isolated and purified from Agkistrodon acutus venom by DEAE-Sepharose CL-6B and Sephadex G-75 chromatography, and the effect of the two on the blood coagulation system indexes was observed through in vivo experiments. Results: A single component of thrombin-like and fibrinolytic enzymes were isolated from Agkistrodon acutus venom, respectively, Their relative molecular weights are 39300 and 26600, respectively. In vivo experiments have proved that both thrombin-like and fibrinolytic enzymes from Agkistrodon acutus venom can significantly prolong the whole blood coagulation time, activated partial prothrombin time, thrombin time and prothrombin time, and reduce the content of fibrinogen, but the role of thrombin-like enzymes is stronger, while fibrinolytic enzymes only show anticoagulant effect at large doses, Conclusion: Thrombin-like enzyme and fibrinolytic enzyme from Agkistrodon acutus venom have effects on the blood coagulation system in animals, and the combination of the two has obvious anticoagulant effect
