Exploiting toxic animal medicinal materials, innovating drug molecular resources and revealing the pharmacological and pharmacodynamic mechanism of traditional Chinese medicine rely on the discipline group of natural drug functional proteomics of Kunming Institute of Zoology, Chinese Academy of Sciences

Because traditional Chinese medicine believes that it belongs to the category of “flesh and blood sentient goods”, it is highly valued. Traditional Chinese medicine for toxic animals plays an important role in the research and utilization of traditional medicines. However, at present, there is little research on the specific active components and mechanism of action of most toxic animal traditional Chinese medicine in China. The main bottleneck is that its components are complex, difficult to separate and purify, and difficult to identify its structure. Lai Ren, Xiong Yuliang, Zhang Yun, Xiao Changhua, Wang Wanyu and other members of the research team of Kunming Institute of Zoology, Chinese Academy of Sciences have long studied the material basis and mechanism of the efficacy of traditional Chinese medicine for toxic animals, established a corresponding active molecular resource library, developed many innovative drugs, and gradually established a “targeted mining technology system of functional components of traditional Chinese medicine for toxic animals based on biological survival strategies”. The characteristics of this technology system are: 1) theoretical innovation: taking the survival strategy of toxic animals as the theoretical guidance to explore the functional components; 2) Technical innovation: proteomics combined with pharmacology is used to track the separation and purification of functional components; 3) Integrated innovation: create a targeted mining technology system for functional components of toxic animals and traditional Chinese medicines based on biological survival strategies, identify the material basis for their survival strategies, and reveal the material basis and functional mechanism of such traditional Chinese medicines. Through this technical system, they identified pain relief, hemostasis, antithrombotic, hypotensive, anti-cancer, anti-bacterial, anti-oxidation, anti rheumatic arthritis Immune regulation and other relevant active molecules reveal the material basis for these medicinal materials to exert their efficacy, and directly prove the effectiveness of this kind of traditional Chinese medicine at the molecular level; At the same time, it also identified some substances that cause allergic, bleeding and other related side effects, providing guidance for the safe use of these medicinal materials. This series of work has laid a solid foundation for the modernization of traditional Chinese medicine and the research and development of innovative drugs for this kind of drugs. It has obtained 30 invention patents and 1 utility model patent, which has produced good scientific, social and economic value, mainly reflected in: 1) revealing the specific effective ingredients of these traditional Chinese medicines at the molecular level. They have identified more than 800 functional molecules from these medicinal materials (including antimicrobial peptides, bradykinin, tachykinin, antithrombotic peptides, protease inhibitors, proteases, bombesin, antioxidant peptides, immunosuppressants, phospholipase, melittin, neurotoxic and skin repair peptides, etc.), and analyzed their structures, action targets and mechanisms; 2) The identification of effective molecular groups provides a scientific basis for the formulation, processing and application of these standards for traditional Chinese medicine; 3) Identifying the substances with toxic side effects such as allergens in these Chinese herbal medicines provides a method for separating and removing the toxic and side effects in these Chinese herbal medicines, and provides ideas for the diagnosis and prevention of such Chinese herbal poisoning; 4) Some active molecules derived from animal toxins have been developed into clinical drugs or are under development, including cobra polypeptide neurotoxin polypeptide, agkistrodon acutus venom thrombin, vespid polypeptide, gadfly anti laren, born in 1972, researcher and doctoral supervisor of Kunming Institute of Zoology, Chinese Academy of Sciences, winner of National Science Fund for Distinguished Young Scholars, and introduced talents in the “Hundred Talents Program” of Chinese Academy of Sciences in 2004. As of January 2014, 125 SCI papers have been published as the first or corresponding author, such as Proc Natl Acad Sci, Mol Cell Proteomics, Hypertension, etc; Invited to serve as deputy editor of J Venom Res; It has applied for more than 70 invention patents. It has undertaken the key programs of the National Natural Science Foundation of China, the National Science Fund for Distinguished Young Scholars, the 973 Program of the Ministry of Science and Technology, the major new drug discovery program, and the directional programs of the Chinese Academy of Sciences. It has successively won the National Technological Invention Award (2013, ranking first), China Youth Science and Technology Award (2011), Tan Jiazhen Life Science Award (2010) and other honors.
